You can not skip Blighttown, but you can make it significantly shorter by taking the shortcut through the Valley of Drakes. A trek into Darkroot Garden is not necessary to advance the story at this point, but it does provide a great spot to level up and tackle a boss before heading into the far more deadly Depths.
